Would these champions make use of; or is it abuse cases if these champions use cotc?

I've been wondering about champions like Diana who either go full bursty ap, which they take thunderlords, then theres the dianas that do items like item 3027 item 3115 item 3116 etc. as a Courage of the colossus user, she goes from a burst mage style to an ap bruiser/fighter with more survivability granted to her kit.

The same can go for Kled who can also do something similar with item 3071 item 3748 item 3022 style items to get the keystone and still do his diver style of play.

Why would it be an abuse case? The keystone has already been nerfed to near non-viability on anything other than pure utility/CC champions. But to answer the first part; Yes, they're able to make use of the keystone shield.

It's still good on singed [slayer-pantheon-thumbs]

it's really not, you're better off stacking DoT with DFT over a shield that wears off before the enemy is able to retaliate the flip.

when going into a team fight you flip one to get the sheild and lay out poison so you can make it out alive with minimal damage

In Diana's case it would actually fix the problem of her failing to be a bruiser and thus has to play as an assassin.

On Kled it could work, but honestly I feel he should focus more on damage keystones like TLD.

But she plays much better as an assassin even if a tanky build was viable

The thing is that she's not supposed to play as an assassin. She's supposed to be an AP bruiser.

She's a classic example of a failed champion. But not an extreme one.
