So, would a co-op game with friends count? Or does it have to be against people? I'm finding contradictory answers.

It has to be a "matchmade" game. "Matchmade" is any time you enter a queue, which only rules out Custom matches. Vs. AI, TT, ARAM, they all count as long as it's not a custom match.
