What do you think about their current states? By not in the update, i'm thinking about those one as primary or secondary assassin, like Master Yi, Nocturne, Kassadin, twitch, tryndamere.
Do you think they need some change, or they are currently healthy?

kassadin and twitch are fine
nocturne needs some fine tuning
twitch is being updated.
his w was reworked to a lingering poison cloud that builds up stacks of his passive per second on enemies, and slows. his stealth mechanic changed, and i think his ult too. it even gets its original name back. "spray and pray"
nocturne is probably being reclassified as a dive fighter, and adressed when riot gets around to a diver fighter update. theyre still debating if vanguards or dive fighters are next for a class update.
yi is considerd a skirmisher i think, but his kit doesnt really fit the assassin description. imo hes closer to a dive fighter than an assassin, even on a squishy crit build.
hes the archetypical melee adc, and entirely a thing of his own. plus, his kit doest have the issues that theyre currently adressing with the assassins. he doesnt burst enemies too fast with too little counterplay, and he isnt a stealth champion.
kassadins kit is still broken, but either riot is planning on giving him a larger rework that goes beyond the scope of the class update, or they just still have no fucking clue how to fix him without changing his identity and completely removing his iconic ult.
Primary and secondary roles are really outdated ever since this happened: http://na.leagueoflegends.com/en/news/game-updates/gameplay/dev-blog-classes-subclasses Yi is not an assassin but a skirmisher, coming from a Yi main.
Anyway, since you were asking I might aswell share my opinion on him.
Youmuu's lost its attack speed which means that it now sucks on Yi. I really like the Maw update though, getting this, Bloodrazor and Black Cleaver for a core gives you a lot of tankiness, more CDR for alphaing and getting Highlander more often and it gives you a healthy mix of AD and on-hit. Plus, the penetration isn't wasted on his E anymore.
New jungle seems like it's going to benefit powerfarming a lot which means basically wet farm dream for Yi in the early game, more EXP and gold. Gonna miss krugs buff though, kept you extremely healthy with attack speed runes, masteries and double strikes.
