In Forest Hills, Queens, New York, Midtown High School student Peter Benjamin Parker is a science-whiz orphan living with his Uncle Ben and Aunt May. He is bitten by a radioactive spider (erroneously classified as an insect in the panel) at a science exhibit and "acquires the agility and proportionate strength of an arachnid”. Along with super strength, Parker gains the ability to adhere to walls and ceilings. Through his native knack for science, he develops a gadget that lets him fire adhesive webbing of his own design through small, wrist-mounted barrels. Initially seeking to capitalize on his new abilities, Parker dons a costume and, as "Spider-Man", becomes a novelty television star. However, "He blithely ignores the chance to stop a fleeing thief, [and] his indifference ironically catches up with him when the same criminal later robs and kills his Uncle Ben." Spider-Man tracks and subdues the killer and learns, in the story's next-to-last caption, "With great power there must also come—great responsibility!"
Despite his superpowers, Parker struggles to help his widowed aunt pay rent, is taunted by his peers—particularly football star Flash Thompson—and, as Spider-Man, engenders the editorial wrath of newspaper publisher J. Jonah Jameson. As he battles his enemies for the first time, Parker finds juggling his personal life and costumed adventures difficult. In time, Peter graduates from high school, and enrolls at Empire State University where he meets roommate and best friend Harry Osborn, and girlfriend Gwen Stacy, and Aunt May introduces him to Mary Jane Watson. As Peter deals with Harry's drug problems, and Harry's father is revealed to be Spider-Man's nemesis the Green Goblin, Peter even attempts to give up his costumed identity for a while. Gwen Stacy's father, New York City Police detective captain George Stacy is accidentally killed during a battle between Spider-Man and Doctor Octopus.
In a battle shortly after, the Green Goblin throws Gwen Stacy from a tower off a bridge in New York. She dies during Spider-Man's rescue attempt; a note on the letters page states: "It saddens us to say that the whiplash effect she underwent when Spidey's webbing stopped her so suddenly was, in fact, what killed her.” Not long after, the Goblin appears to kill himself accidentally in the ensuing battle with Spider-Man.
Working through his grief, Parker eventually develops tentative feelings toward Watson, and the two "become confidants rather than lovers". A romantic relationship eventually develops, with Parker proposing to her and being turned down an issue later. Parker went on to graduate from college and becomes involved with the shy Debra Whitman and the extroverted, flirtatious costumed thief Felicia Hardy, the Black Cat.

Spider-Man has a supernatural ability to detect threats in his general vicinity, having his Spidey-Sense detect it. Enemy champion who are Camouflaged are detected an additional 100 / 200 / 300 / 400 units away than its normal detection radius (at levels 1, 6, 11, and 16). Enemy champions in Stealth will be detected at 200 / 300 / 400 / 500 distance away sourcing from Spider-Man. Lastly, enemy champions who fire projectiles in a large radius around him (1100) are detected.
When detecting an invisible enemy champion or a fired projectile from enemy champions champion using Spidey-Sense, he immediately reduces all of his cooldowns by .5 / 1.0 / 1.5 / 2.0 seconds. This effect has a 8 / 7 / 6 / 5 second cooldown per enemy champion but resets immediately if he loses vision of them. He does not need to have sight of an enemy champion firing a projectile to gain this bonus.
- Spider-Man “detecting” a camouflaged, in stealth, or unseen projectile firing enemy champions does not reveal them and make them visible. It does however, show the visuals as depicted above in the picture around his head, pointing in the direction of the invisible enemy champion within 30° of their current direction.
- Spider-Man's Base vision radius is 1100.

Spider-Man shoots out a concentrated glob of his webbing, hitting the first enemy unit in its path, dealing 35 / 55 / 75 / 95 / 115 (+40% AD) in physical damage. Damaging an unit will slow them for by 15% / 20% / 25% / 30% / 35% for 1.0 / 1.25 / 1.5 / 1.75 / 2.0 seconds. If the damaged enemy unit is debuffed by any slow effect, they will be stunned for .5 / .75 / 1.0 / 1.25 / 1.5 seconds instead.
If the webbing travels to its destination without damaging an enemy unit, it creates a small area on the ground, lasting for 3.0 seconds, where it would apply only a non damaging slow for the same duration, or applying a stun to a slowed enemies, destroying the webbing to do so in either case.

Passive Critical Strikes against enemy units who are slowed or stunned deal an additional 30% / 35% / 40% / 45% / 50% critical strike damage.
Active Spider-Man reflexes are unmatched, preparing himself for the next assault against him. He dodges the next single auto attack within 2.0 / 2.25 / 2.5 / 2.75 / 3.0 seconds. If he dodges an auto attack this way, his next 2 auto attacks will critically strike within the next 3.0 seconds.

Spider-Man shots out a long string of Webbing high up, using it to help him dash feet first in a direction, knocking aside minions as he goes. Spider-Man can traverse over terrain as he dashes. He may also land on top of terrain as a final destination, and wall crawls on top of it for 3.0 seconds. If he does not cast a spell or auto attack by the end of that duration he drops down near the closest available location.
As he dashes, he gains an additional 200 / 250 / 300 / 350 / 400 auto attack and spell range, including increased range on the second cast of Web Sling, and can interrupt the dash with an attack or spell command. If he lands on terrain, he gains the same increased auto attack or spell range for his next attack or spell as he wall crawls.

Spider-Man does a high speed dash, damaging all units he passes through for 80 / 150 / 220 (+20% AD) in physical damage. If he collides with terrain at any point in the dash, he stick himself to the side of it for up to 1.25 seconds. In this time he may recast Maximum Spider without cost, reducing the range by 200 but increasing his dash speed by 200. He may recast this ability a maximum of 4 times, granted he collides with terrain each time.
