A: poison-master assassin (assassin) Dot Assassin sounds interesting. If you manage to make a champion that even when ahead still relies on his Dot instead of bursting with another ability (Teemo Q, Casseopia E), it would result in a very interesting playstyle.
B: earthworm monster (tank)
Since the burrow mechanic has already been used
the only interesting aspect I could potentially think of is a giant champion that consist of multiple parts (which is really hard to balance), maybe you could elaborate a bit more.

A Fullmetal
and the T1000 come to mind. I do like the concept of an assassin support though. (high mobility CC or Utility and Burst ?)
D: jade monster (tank/support) Don't really have a clear theme in mind from reading that... Does Jade have an assoziated power ?
E: half-giant (tank/fighter)
Does have a lot of competetition from other giant champions
and might need some defining features.
Since some stories feature gaints as children of the earth maybe he could be a terraformer that does more than place a single wall with an ability.
