If he actually is just trying the game, there's no point in him having to deal with masteries/runes yet, a 10 Champions pool a week should be more than enough for him to try out things and why are you letting him play agaisnt lvl 30 Silver/Gold people as a newbie?

That falls on you, and how you teach him how to play.
THere is zero reason to even use the runes and mysteries if you can't use your champions. Since we have over 120 Champs... Maybe get off your high horse and direct your sibling to develop their own play style.
They don't ban an account just for playing from multiple IP locations. If that were true, I'd've been banned years ago: 2 different personal laptops at multiple locations (tried on a third, but found out it was a toaster after 1 game), I've used parents' desktop when I go home, I've used multiple computers at an Internet cafe... yeah, if it was strictly "New IP, BAN!!!", I'd be done.
Not too long ago my brother got me to install and try League. So since I remember getting into League as a complete noob (Still a noob but you know what I mean.)
You may not like to hear this but it's probably best to start at ground 0. Bots are fine but if you move into PvP in duos you really will skew his perception on the difficulty of the game. I was starting to get good at bots so go into Regular mode and get crushed cause my brothers level 30 vs my level 11 gave me a higher level lane opponent. Learning support helps mitigate this a little bit. But in PvP he is best off in SoloQ or from you smurfing, possibly ARAM....
Champion pool: The Pre Level 6 Champions are DIFFERENT than the free champion rotation. Garen is included though a free champ from liking FB or Following on Twitter or something. Tristana and Alistair are also free. Don't tell him to start playing random champions you think are OP. Let him identify with the champions they enjoy but start with Garen, Ashe and maybe Lux (I loved Lux when I first started) or Ryze for Mid. He'll learn a few roles and they are relatively easy to pick up at first. When my brother would tell me "Pick this guy, he OP." and I would hate it, but then grab Poppy and LOVE it.
And in response to last hitting and the lack of Runes and Masteries... This is fine. It's important to learn these things with lesser power and grow into it. It's hard but gets easier as your skill increases as well as your level. You don't throw a 16 year old into a Lambo to learn to drive, they'll spend all day looking for the windshield wiper switch. You give them a Corolla and let them grow into a Lambo.

so wait, you are admitting to Violating the Terms of Use according to III C in the http://na.leagueoflegends.com/en/legal/termsofuse
it clearly states: "C. Login Credentials. In creating an Account, you will be required to select a unique username and password (collectively, “Login Credentials”), which you will use each time you access the Game. You may not share your Account with anyone other than as expressly set forth herein, and you are entirely responsible for maintaining the confidentiality of your Login Credentials and for any and all activities (including purchases and charges, as applicable) that are conducted through your Account. Please notify Riot Games immediately if you become aware of any breach of security, including any loss, theft or unauthorized disclosure of your Login Credentials."
When you installed the game you agreed to this. If you violate it Riot can just perma your account.
If you really want to teach your brother the game, level a smurf and play botgames together. Buy him the champ you think suits his way to play/personality best and teach him how to play it / build paths and for the love of god, teach him reactive build paths. You do Him and Yourself no favors by violating ToS
i would let him try out some of your champions, try to let him get a grip on what role he likes and if he even likes the game, then if he does hopefully he would have a grip on what roles and champions he wants to focus on learning. but yes he should eventually make his own account

Well its how you started the game, right? There's a learning curve to MOBAs and your brother will have to take the small beginner steps to get to the better stuff later. That's why the leveling system exists, so that people can learn the game mechanics.
Yeah I'd rather not give him champions like Kalista or Riven when he's new to the game. Let him stick to Ashe :)
He's in luck then, Garen is also on free rotation this week. Problem solved :)

I am not entirely sure where you are going here. Yes, new players have to make new accounts. This is nothing new in the history of gaming, so...??????? What exactly are we discussing here, it seems we've pretty much covered it.
