What is S rank based off of for support?
It's not assists. The few times I get S on a sup is when I finish kills cuz my ADC died first. I have heard that roams help but I have done this and still only gotten an A. What is the trick?

At least 20+ assists for S- . Cs. Wards.
Wards. CS. Kill participation. Kda. The champion. The role you're playing. Your elo. How other people play that champion.
Got S- as Janna last night. Went 0/1/20+. Single digit CS. Don't know how many wards. If damage shielded is a stat, then I think I did OK with that during lane phase. Don't know what else can contribute.

Don't believe people when they say kda, cs, wards, ect affect your grade, its unlikely to be true.

You get compared to other people in your region who played the same champion as you in the same role as you, the higher your score amongst these people the better your grade will be. https://support.riotgames.com/hc/en-us/articles/204211284-Champion-Mastery-Guide
Also, there are a lot of people giving false advice, a lot of people believe that kda and cs is somehow a direct factor in your grade. Riot has also noted that even the best of players cannot consistently get the highest grades (though they do get them more often), which leads me to believe there is at least some degree of luck involved.
While that example game wasn't a support, and I don't have any example support games to give you, I doubt warding as a stat counts for mastery grading, just like kda and cs are unlikely to be a direct factor for other roles. If there even is a difference between the roles and how they are graded.
There really is no trick, unless you count being good at the game and getting lucky as being a trick.
It's not going to be any one thing and they are going to be different depending on who you play. Things like damage tanked, damage dealt, damage healed, damage shielded are obvious possibilities for support in general.
Those specifics don't matter however. If you get a good game where you are playing generally well, you do things that will qualify for an S grade.
