I have 3-5 really off meta builds that I use pretty often

Revival Time -> Zombie Sion Full A/S and magic pen -> AP Varus Full magic pen -> Mordekaiser/Teemo Full scaling AP -> Moses Janna 30 scaling mana regen -> Sona

Probably not.

It seems similar to the way that the old glyph system, or current talent system, in world of warcraft functioned. They sacrifice a gradient of many different niche builds, for only options which support the "tried and true" builds.

They rarely support hybridization of two different builds and generally just act as a means to pigeonhole players into specific builds, which allow for better manipulation of balance for the devs.

It worked out great for world of warcraft though... http://imgur.com/f0757NU

Right around the start of cataclysm, they started doing things like "Once a talent specialization has been chosen, players may only place points in the primary tree until at least 31 points have been spent there." Which would later change into, "we've gotten rid of the 'fluff' talents and are only going to give you the choice between cool and impactful talents. All of those number things are just now integrated into everything for you instead." It wasn't just the talent system though, they also started supporting optimal secondary stats based on specs, and other things which follow the same pattern of "we will help to prevent you from not playing correctly".

Though, it seemed to work out fine in that situation, league can't fail, because it's not suppose to.

Hopefully they won't affect anything too much. I would be super disappointed in one of my favorite games if I couldn't play AP varus' anymore.

From what I've seen of them it doesn't really look like that'll be the case.
