Well Brand's new intro paragraph alludes to such:
Once a tribesman of the icy Freljord named Kegan Rodhe, the creature known as Brand is a lesson in the temptation of greater power. Seeking one of the legendary World Runes, Kegan betrayed his companions and seized it for himself—and, in an instant, the man was no more. His soul burned away, his body a vessel of living flame, Brand now roams Valoran in search of other Runes, swearing revenge for wrongs he could never possibly have suffered in a dozen mortal lifetimes.
And then Zoe comments on world runes in "Meet Zoe":
As Zoe stepped out of reality, she wondered if the new crater would lead some mortals to find the World Rune that was nearby. The tall girl or that interesting boy might even be the ones to discover it.
It would probably be funny if they did, she decided.
So Ryze and Brand are both after runes and Targon seems to be very aware of World Runes and their importance, considering they sent their cosmic messenger girl to make one easier to find for the mortals in the area. The first point makes some sense since Brand and Ryze have been listed as rivals on the main League site for years, so there were likely always plans like this in mind, or at least some direction for brand involving World Runes, and the second point just raises more questions about Targon and how involved they actually are in Runeterra.
