Realistically speaking, it's the map. Each position offers it's benefits, and certain classes of champion can make use of those benefits better than others.

Taking 1 and only 1 Jungler and 1 duo lane offers the best gold/xp distribution among the team. Mid lane offers a lot of global pressure for anyone with the range or roam to make use of it. (Why mages and assassins usually go mid) It's also the shortest lane, making it safer for those glass cannons. Marksmen can survive sharing XP with the support, because they rely more on auto attacks than abilities.

And while you can justify very specific breaches in this kinda setup, you really can't break the formation with out really changing the nature of the map itself.

Because of the map the dragon in on the bottom and easy to solo so two players take control of the lane and the minions reach mid fastest

It's actually many factors that come into play. Mid for ex is the shortest lane so squishy apcs can get the most farm and have easy access to their turret since most don't have an escape. If they were to go not or too they would get less farm and have a longer trip to tower. Dragon decideds the other two lanes and why would you want your adc all by themselves? That role literally needs another player to carry them to late game to be releveant.
