Obviously there are new champions first and foremost.

Furthermore if you missed a lot of Scuttle Changes. Here are a list of Jungle Routes that you should probably learn, here are the new routes that are about to come up in the new Season. Even if you're not a Jungle player, you should really learn these to understand Gank Routes and how different Junglers have different goals. The Scuttle Camp is basically worth a Gromp nowadays, and farming as a whole is a lot less valuable, so you need to help your Jungler win their early game goals so that they're useful as the game goes on.

We're also approaching a new season, so everyone's kinda relearning the game soon anyway. Just look up the newest Preseason Changes for that.

Mid 2016 (Season 6)? It's been a while, I'll see if I can't try to list some of the stuff that's changed/added (sorry in advance for the big wall of text).

Runes & Masteries have been simplified. Instead of purchasing Runes with Blue Essence or RP(IP/Influence Points have been replaced with Blue Essence, which can be earned via Missions, FotD, Honor System & Disenchanting Champion Shards in Hextech Crafting), you unlock them by leveling up your account. Each Rune Tree comes with atleast 3 Keystones and while you're limited to one Keystone per page, these particular runes open up different playstyles (Glacial Augment giving you AoE Slows & a free Frozen Mallet proc on your auto attacks & Electrocute giving you a small burst of damage after successfully hitting 3 consecutive attacks).
New Modes have been tested out or added to the game. This includes a Sandbox/Practice mode for when you want to practice certain champion combos & League's newest mode Teamfight Tactics (League Autochess). There was also Nexus Blitz, which was a fast paced gamemode with Mini-games scattered throughout each of the matches, hopefully this returns very soon.
The Honor System has been reworked. Through the New Honor system, you can get all sorts of stuff like Key Fragnments and even Medieval Twitch or Gray Warwick.
