No, she doesn't have life essence anymore; she's dead. It is probably the power of his lantern. Thresh is undead by the Necromagic of the Shadow Isles. I'm sure that Flay and The Box is also part of his power.
Thresh seems to become stronger the more souls he gathers,but that might be just a gameplay mechanic
He most likely imprisons and tortures them to break their will and turn them into soldiers of the shadow isles,thats why he seeks stronger souls(or he simply enjoys imprisoning and torturing them)
