It's just a thing he puts on his face, like using lipsticks or whatever? It shouldn't come as that big of a surprise. It seems only Classic and Nottingham have those markings.
You shouldn't be asking whether if he is human or not considering League has been saying that these 
are/were human.
He adds another for every man he kills.
It's how he uses his moon crystal magic.
But seriously, I don't have the faintest clue.
Maybe they're some sort of athletic Eye Black paint. Since he's an explorer it would make sense.
Fangs, obviously, they have significance in japanese culture where having said triangular marks in ones face means that you're wild and don't follow rules, see princess mononoke, Kiba inuzuka or Jio freed.
