I'm looking through some of the champion bios, and I find Vayne's section. She had her lore update, but I'm not sure if these related champions section has been updated as well.
Im sure it would be confusing to the Layman people reading her profile page, but truth is it's just a joke reference. You got your Dracula, Wolfman and Frankenstein while Vayne is Van Helsing.
I will admit there are other examples of outdated relations. Such as Urgot & Sion, like fucking hell there would be a hundred years or so difference between the two lifetimes. And its not like the writers wouldn't have known that impossibility at the time of Sion's update.
Perhaps it's not up to the writers to chose who is apparently related to who?
