I listened to Xayah's voice over and one her lines left the impression that there are real vampires unlike 
I always wondered if vampires existed in Runeterra just to know how they would react to meeting Vladimir. Would they want to eat/drink him? Are his powers useless against them (like trying to drown a fish?). Would they fear/respect/understand each other or what? Could he even be well known among vampires?

I'll just say this bit: We are allowed knowledge of what the powers at be are willing to tell us. People are just now accepting the overall possibility of aliens existing.
Honestly, there could be a plethora of things we believe are made up or fairytale that could have very well existed. Afterall, if you keep a child secluded from the rest of the world and hand her an stuffed elephant plushie, she'll believe that such a creature is made up and is not real. Especially if you tell them that.
