Autofilling a support to top-lane or jungle is generally a bad idea, can you imagine locking in top as a top main and getting it when suddenly the support decides he will only play top, forcing you to play support? Babysitting another champion instead of afk farming? Autofill basically does just that, it forces you to potentially play roles that you deem either "not fun" or that you are bad at which in turn makes your allies not have fun. Games are used for entertainment, so the entire idea of striping someone of their fun goes against the VERY nature of gaming. It's like a dodge(lose) or lose situation, a big "F you".
Yes it is a GOOD IDEA to find a few champs in each role that you enjoy playing, but it definitely shouldn't be required. Especially since someone could ban or take your champ for that role.
The concept behind autofill is inherently dumb, faster queue times for worse quality games is a bad trade-off. a manual fill option makes sense for people who honestly don't care, but that's it.
Maybe even having it ask after waiting in queue for a few minutes if you WANT to play that position? Like "Found a game missing Jungle position, would you like to play Jungle or continue waiting?"
I'll Play | Keep Waiting
Which would go away after like 5 seconds, or if someone else queued up and took the spot.
I generally go Mid or Sup as
