How do Towers know who to target?
Like, in the lore.
Do they magically know who is an enemy? If somebody truly in their heart decides to join the winning team, does that mean they can bypass towers?

They were programmed by Viktor to only target the inferior flesh species.
On a more serious note I imagine it's something to do with the nexus summoners.
I'm going to disregard Riot's retarded lore erasure, as if I don't then there is no explanation.
At the start of a League match, when the summoners on each side spawn their champions (or for the lesser summoners, the minions), they spawn through their respective Nexus' (Nexi?). This leaves a distinct magical trace, like a fingerprint, and enemy turrets will lock onto that. Summoners skilled enough to be assigned champions are also good enough to suppress that trace, so that turrets will not notice champions unless there are no minions to mask them or they draw attention to themselves by attacking an enemy champion.
