If I'm not mistaken the average Nasus player has 300 stacks by the 20 min mark. So anything above that is a good place to start.

If you farm with q perfectly, that's like 20 stacks per wave. At 20 minutes, that's like 740. (You miss about 3 waves in the beginning of the game). If you can make at least 50% of that by the 20 minute mark, you'really pretty much golden. Make some benchmarks like trying to consistently stack 250 by 20, 300 by 20, 350 by 20, and then 370+ by 20. Once you get to that last one, if you'really consistent, you're well above the average nasus player and can do basically whatever you want.

Focus people with low armor. Your e is flat penetration and multiplies the hell out of your damage against those that do not stack armor.

Okay, since I see a lot of Bad Nassi, I'm gonna put down a brief "How to not piss off your team while still being Nasus" guide.

1: Stacking is important, but not as important as objectives or living teammates.

You should NEVER focus on stacking when the enemy team is pushing a lane that you can defend. Same goes for teamfights. You bring a lot of power to teamfights and if you're not present for that fight your team is going to suffer and likely lose a 4v5. Then you'll lose a tower or two, and the enemy team will likely dragon, baron, or snowball into victory. Then you'll get flamed and reported.

2: Eventually, you have to push.

It's every Nassi's dream to go and achieve the legendary 1000 stack "Nuclear Stick/Pimp Cane." situation. However, unless you've got at least an hour or more of freefarming where you hit every minion, your not going to get those 1000 stacks. You need to be constantly thinking to yourself: "Should I keep farming stacks? Is it time to push? Does my team need me for an objective/fight?"

I've seen Nasus players attempt to justify them never taking toplane towers, and I've never been convinced. My personal motto is that when a tower falls or a dragon gets taken it's time to start pushing them. Doubly so if it was one of your towers since you need to even up the tower count sooner rather than later.

After 20 Minutes, take the Minute you're in and multiply it by 20. If you're above that stack-wise you're good to go. Before that, having >130 at 10 minutes is great. Your whole tactic for the game is split pushing, but be smart about it. You're a 1v2 monster after some point, and they either need to be very far ahead or send 3 people to stop you. Also you obliterate towers. I play Nasus occasionally, and that's how I play him. He's pretty good until mid to high plat where people understand that to beat Nasus you have to ignore him and 5v4 destroy his team. Have fun!

Also if an enemy places item 3512 don't kill it. It's a free stack dispenser! ;)

If you are new to Nasus, I would recommend that you start fighting your lane opponent when you have 200 stacks and Sheen in most situations. This would typically be around 13-17 minutes into the game, so your normal build would be either Frozen Heart and Sheen or Spirit Visage and Sheen.

Nasus is strongest in the mid-game, so you want to take this time to pressure the map. Whether you do this by grouping to pressure objectives or split-pushing to draw multiple enemies up to your lane is up to you.

Honestly, I'd say map pressure is probably more important than building your Q stacks beyond a certain point. In many situations, I'll just AoE a wave using Spirit Fire and take whatever Q-stacks I get rather than Q each minion to get it under a tower more quickly. This is especially true in lower ELOs where your team is more likely to get caught out doing something stupid without you (with no wards nearby for TP).
