I've been endeavouring to master 
. I've watched pro streams, read dedicated mains guides, and played him a lot, but with season 6, I'm confused as to which build I should go.
Hell I've heard Guinsoo's is great on him from some people!
Any ideas?
Well I'm not really great at twitch, and I've tried the same thing as you
After a bit of trial I decided on the build
With summonses 
and 
I'm not really great at twitch so if u don't like my build u should change it
The botr is for early life steal and dmg
The boots are for speed basically with the atk speed a bonus
The bloodthirster is for the life steal and high dog
The runaan hurricane is for attack speed and critical strike - by this point twitch should be in his strong form
The infinity edge is for the damage
And finally the green sword thing is like quicksilver sash just with ad
Pre lvl4 twitch is really strong, after four he can't really hold his own
By the time u get runaanshurricane he should have reached the level in which he can assassinate an enemy
i am assuming you are talking about lane twitch and not jungle twitch. The BOTRK nerf hit him hard but you will still want to start with. That and then ghostblade right after is the must standard build. Once you get those two assuming you are not behind you should be able to win any duel. Twitch exales at catching people out so anytime someones goes to push a lane to farm by themself mid game you can gib them. BOTRK and GB still give you them best way to burst someone from stealth.
from my experience, you do a rush similar to that of tristana and cait. Meaning, you go bf into runaans. After that finish ie, and then the 3rd item depends. It can be either static, bt, or ldr depending on the situation. After that you just build the remaining items. Sometimes though, if I have to back early and I don't have enough bf, but I have enough for a long sword or two, I will get ghostblade first. Bork isn't as good on twitch as it used to be due to the nerf it got.
