Noob Question
While playing as Vayne does her passive always work while moving towards an enemy? What if they are out of vision? Or stealthed? Can I use her passive to tell if an enemy is in our jungle before I cut through it?

No.
Only works on nearby visible enemy champions, so it doesn't work if they are stealthed (Twitch, Rengar, Eve, Shaco etc.) or in the fog of war (like in jungle when you don't have vision on it, or even in bushes).
But you can tell that enemy Vayne sees you if you are in the bush by checking on her status bar (if she have the passive buff then she sees you). Though, I am not 100% sure if it actually shows up there... If no then you would need to look at her ms.
