Honestly, we have no idea how Void reproduce or even if they do (Kog's "daddy" doesn't mean he was spawned by him, necessarily). Rek'sai could only be a queen in the sense that she is the largest and most commanding of her species in an area. She could also be able to create more. Void biology seems to be highly unexplored so far, so there's no definitive answer other than a "maybe".
I remember a Rioter saying she's the largest specimen of her species, and her passive says she's a "queen". I'm not sure.
