Not with base rules but I can't see why fan made (or future expansions) rules could allow of PvP or at least team v team.

The original design was competitive. I'm unsure if it was competitive in that each person was trying to kill as many minions as possible or if it was about hurting each other. But about half way (or more) into the design of the game they shifted to pure co-op.
