Is it possible that someone at level 30 doesn't know what jungling is? I just finished a game in twisted treeline where we had an ekko jg. At first he didn't buy talisman or machete and so I was like aight w/e he must've forgotten or something. I asked him about it and he didn't even know what these items were. He also didn't recognize diana(as in he didn't know her name). He said he played mostly bots getting to level 30. So, do you think he was just trolling me super hard, or is this actually possible?

I mostly played bots to 30. Had to fill adc in my 1st ranked game, went full tank Ashe. Got carried. He may have been new or him trolling is not impossible as well.

There's a limited bot pool, so it's possible that you never encounter Diana if you only played PvE matches. And tutorial is pretty bad so it's possible you never end up learning what jungling is.
