Will autofill ever get removed?
I really don't think it would be bad. Queue times would still be fine and i wouldn't have a zed support because mid didn't let them go mid. This also makes less games coinflip if everyone is on role.

No. This comes up constantly as a question; autofill is mandatory for the game to function and will not go away.
It's unlikely. We used to have a queue type where every single person could choose the exact role and champion they wanted to play and could choose to avoid teams with obvious trolls before even entering the queue to find a game. It was extremely successful, so it had to be replaced. I suspect the reason it was removed is because it caused a huge drop in ranked participation, but I have no data to back that up.
You want autofill to go away? Play support more often. Problem solved.
Autofill isn't the root problem instead the problem is the role of support. It has always been this way, but due to the pro scene Riot will never remove the role ergo autofill must remain.
