Instead of adding the 6th player, he would be shotcaller/coach. He could use F1-5 to cycle all players, ping, advise ganks, and instead of abilities he could have arrow waypointer, target marking enemy champions, impersonated shop buy calls. Instead of mini version tab he gets one where he can see gold and exp of all teammates.

Alternatively, (s)he can be courier. Meaning that (s)he has the ability to carry item(s) to players and maybe even have a trinklet slot. In lategame when all are full build he can carry vision wards for team.

I don't really like the idea of the courier, I don't see why anyone would want to play as a postman in LoL.

However, I love the idea of the "shotcaller/coach". I always liked it when team games give one player the opportunity to be a "commander", like in the battlefield games, and I loved the concept of Abathur in Heroes of the Storm (for those who don't know, it's basically that : a very squishy hero that cannot fight himself, but has global abilities and thus can lead his team from afar).

A player which can focus exclusively on macro would make the game much more strategic and feel more like a chess game than an aram.

So yeah, for what it's worth, I fully support the idea !

So the 6th player would be playing a strategy game? All cool but wouldn't work if rest of players would just ignore him

You can already do this with a mic and premades. For solo queue? You can be 20/0/20 in game and people still won't listen to you, don't know why you would expect any of these street urchins to listen to a coach. ;)

I think that a dedicated player for strategy / macro play, would be a nice addition to game. However, players should have an option for mute and ignore.

Clearly, there should have a ranking for the 6th player (that is independant of your play ranking) and some of them are much better at macro play than others, some might be able teach players new tricks or are just comedians who help the team keep the sprit up endure game even when they are behind.
