So, how do I know what masteries to choose?
When I watch streams, I notice that they change their masteries in champ select everytime. How do I know what ones to choose?

I think it's pretty straightfoward :|
Read what they do and test different stuff.
I don't watch streams, and rarely watch pro-play. But whenever I choose my masteries I take into consideration what I want out of the champion. What I want to do.
On Sion that I've been playing recently, I get 3 points in utility (1,5% movespeed) (plus 4,5% on runes). Then 21 offense, in defense, since my point is getting to people when I'm in his Zombie passive, I get 15% slow resistance.
On Lucian because he procs Blade and Spell Weaving I get 30 points in offense.
On Aatrox I go 21 offense because he's weak. 9 defense.
Note here that the OP isn't asking for the optimal masteries -- which you get by copying whatever Senpai-san wrote on mobafire -- but instead how himself chooses his masteries.
You choose them by, 1st - Knowing them. So get to know them through reading and experience. (Starting to play LoL I thought 3% increased damage was irrelevant, untill I started noting the difference of having them and not having them). 2nd - What champion are you going to play, and how do you like to play the champion.
I go 21 offense on Soraka, 1,5%movespeed, 15 AD runes and spellthief. So the bananas hurt c:
First decide where you want the points. Generally, if you're an assassin/mage/marksman you want to run 21/9/0 for the damage. If you're a tank/fighter, you want to run 9/21/0. You generally don't want to run utility unless you're a support, but some mana hungry mages can use utility mastery as well.
I'm gonna use Ashe as an example. So you want to take 21/9/0, since you need the damage. Now, go ahead and take everything you can use in the offensive masteries. Basically all AD masteries, attack speed, etc. Now, remove points that you need the least until you have 21. Then go ahead and take 9 points from defense.
