I'm just wondering this because my friend is trying to get tier 7 nautilus and is holding me against my will to play with him every night until he does, so I was wondering if this works, so I can let him have the kills and get tier 7

To increase the chances of getting an S? Yes. Just make sure to farm up and last hit too, but do it when appropriate.

No. Riot specifically set kills to the exact same value as assists in the support role
If you want an S here is the best way to go about it.
Side note, Aim for at least 240 CS a game, have less then 1/4th of your kills in deaths. Attempt to last hit towers.
I main support. Kills means nothing. Multiple times i went with 0 kills but some deaths and many assists and get S. As with Nautilus I found very hard to get S and I realized why - he is classified as tank, not support.

OP if u want a shortcut, you could play 3s blind pick twisted treeline to get that s ranking
why? because some scripting bots have recently mass invaded 3s blind pick normals, there appears to be at least 1 every game and I don't know why riot hasn't disabled hextech drops from the 3s yet.
It depends. I've hit my S's by:
If he wants an S so badly on Nautilus play Nautilus top.
Play him top, you will get more Kills and more cs.
