Ping spikes mid game affecting competitive play

When I am in games my ping will randomly spike, It's only started to do it the past few days, but It's beginning to affect my ranked play, as seen in the Gyazo link, when ping testing I will run around a 44ms ping to chicago. However, while playing league with nothing but Pandora and skype in the background (both are always open and I have always played with 33-45ms ping) recently I have been having frequent spikes to nearly 1-2s ping. I know it isn't my PC to the Router because I have set up QOS protocol so league would be the first thing prioritized, so it's either comcast or riot. I have restarted my router several times through the week, and it hasn't helped, this really is my last resort. has anyone else had similar issues, or know how to help speed the connection up?
