Would they team up?
Would there be any scenario where Kalista would actually accept a team with Thresh and fight for Shadow Isles? If Shadow isles was in theory ever threatened?

Has Thresh betrayed anyone? He's pretty up-front about his evil intentions, so there's no reason for Kalista to hate him. Thresh was not responsible for the Ruination; he's as much a victim as Kalista.
Then again, Kalista has in-game quotes for an enemy Karthus, expressing dislike, even though Karthus hasn't betrayd anyone we know of either. So Kalista isn't best buddies with the other Shadow Islanders.
Unlike Yorick, Kalista does not express any desire to cleanse the Isles and pass on. So if the Isles were threatened, she would probably fight back.
So in short, I think she would fight alongside Thresh in extreme circumstances. But she wouldn't be happy with it.
Possibly, I mean the guy that she really hates from the Shadow Isles is hecarim for obvious reasons.

This is a great question and it's hard to answer. Thresh is really smart and he doesn't necessarily betray people. He might use people for his own gain but if you trust some spooky guy like him who has a lantern full of crying souls it's like ¯_(ツ)_/¯
Now if Thresh was being legit I can see him working together with a lot of people because he isn't a completely unsociable person. Not sure how impulsive Thresh is when it comes to torture but if he isn't, sure he would work with Kalista if he found any kind of enjoyment in it. For example maybe Kalista would give the souls of her victims to Thresh.
Now on Kalista's side, she has a bit less self-control I think and has had the black mist and vengeance eat her up more. I think she only hears the voices of victims of betrayal for the most part and would not ask Thresh for help. If Kalista ever found herself fully maybe she would form a partnership with Thresh. Maybe to fight Hecarim or something, Thresh would only accept if there was anything interesting to get out of it.
Also a major question. Do these shadow isles champions have each other on cell or something? How will they know where each other is at?
