It's a joke playing off the fact her passive only goes to four stacks, she knows full well how to count past 5.
Until about mid 20th century or so, the vast majority of people did not have college level education and in fact most people didn't even make it past highschool; most people sported either primary or middle school level degree, and illiterate population still posed a substantial portion of the total.
Now imagine the level of education people would have in a mostly medieval world where outside of those nobodies on a mountain who has somehow developed god-level technology, and 1 state and their downtown sibling literally just 2 blocks away from them, most of the tech is only advanced up to primitive firearms that still use flintlocks.
I wouldn't be surprised that Annie, who seems to have received little to no education in her entire life so far (given she was very young, used to live in the mountain and spent most of her time playing, and her family died (debatable on whether she indirectly killed or murdered her mother) and so no one to teach her anything from that point on), is not very well-read and academically sophisticated.
Not to mention, in the end of the day, she's still just a child who's less than 10 years old.
As someone who works with kids on a daily basis, some kids I have to wonder if they can even count to five by that age.

If you take the quote seriously you have to consider where she lives. She's not exactly from Piltover where education would be the norm. She's not even from Noxus proper. She lives out in the woods in a not so technologically advanced place.
If you take her quote into context with other quotes like "beaten by a little girl" you can see it's just her taunting others. She's little, but has the power to take down most adults.

Well, she WAS homeschooled. Besides, I always assumed Annie was supposed to be like six based on how she acts.
She can count to ten at the very least, assuming she understands the concept of numbers.
It's just a joke on her passive. It's no different from an adult saying it while breaking your fingers.
