Kalista and Thresh have a few things in common, lore-wise. They're both SI champions that have the ability to take souls. Thresh seems to be able to take souls from his (unwilling) victims, whereas Kalista can only really take your soul if you willingly give it to her. Kalista's souls seem to be bound to her, so I was wondering, what would happen if Thresh killed Kalista? Would he be able to take every soul bound to her?

If Thresh killed Kalista there'd be bigger things to worry about, mainly the resulting explosion of souls and the devastation such an event would cause; you gotta recon if the primary consciousness of Kalista is destroyed, things would go to hell in a hand basket real fast!
On your question though, I'd imagine Thresh has the capacity to take every soul, perhaps not all at once, but a good number of them, of course in the 208+ years since Kalista's death who knows how many folks invoked her. I'd recon that very few of the souls that are a part of Kalista would be worth Thresh's time though, considering his main thing is collecting the souls of the strongest that Valoran has to offer, not just any old plebs. I suppose it'd be up to his own discretion, or perhaps the will of someone higher up the Shadow Isles food chain than him.
I think Thresh is like the Jailor of the damned character, who collects the souls of the deceased. Considering his death animation, he himself is a twisted soul, and collects other souls of the damned using an enchanted lantern. He hungers and thirsts for more souls, even active hunts for victims.
Kalista, on the other hand, is the Embodiment of vengeance. Enough magical energy makes up the fury and hatred of people where her existence can be real. Think of her as all the souls with "unfinished business", hunting for the traitors who killed them/betrayed them in life. This is also why she refers to herself as "We" instead of "I". I imagine that Kalista would be a good snack for Thresh, and the two often play cat and mouse... (Think of how many souls have it out for Thresh for eating them? they want vengeance against him!)
but yeah.... I think they exist from the same dark energies, but are definitely not buddies.
Conspiracy Time! What if souls that want revenge and are consumed by Thresh create shades like Nocturne or Fiddlesticks?!
If Thresh did defeat Kalista I'd would imagine he would be able to gather the souls from her if he acted quickly. Thresh soul sucking power isn't really described to well in lore terms for all we know he can just capture Kalista on the spot since she is a spirit. She may even avoid him I'd imagine some people would love to take revenge upon Thresh (Lucian...).
On that theory: she is undead so the idea of killing her is probably far-fetched (which really says something) However, I'd imagine that she is like on giant conglomerate soul that really has no way of separating. Not sure though.
