It was Zilean. He was one of the original 40 and he controls time so he'd obviously be first.

Hmm ... well, it had to be one of the 20 betas ... unless you're going back to the first game ever played (which would probably have been before the game even went public), where you'd need to talk to Ryze and Tryndamere (the people, not the champions).

You can't get into bot games without playing the tutorial in which you are forced to lock in Ashe.

... But Nami wasn't one of the original champions, so it's a reference to something else as well. Possibly simply a reference to pick order's first pick doing dumb things (the songs old enough I don't know what Nami's state was).
In the First Interns at Riot video they showed alpha footage of the game, and it looked pretty drastically different. They showed Singed and Annie (Annie looked more like a teen then), maybe some others.
Point: the answer is whatever the first champion made was, that's the first one locked in, since that champion would be used to test the map/minions/engine.

I'm taking my bet that is was 1 of 4 champions. Kayle because she was a very iconic Champion back then Or Annie because of how awesome she was but then i think about tryndamer and ryze both being designed from the main founders of the game.
