While reading the guidelines for Memes and Games, I saw:

And it makes you think, who really was the first champion that a player locked in? (insert thinking emoji here)

Like the legend of the first pick He's instalocking Nami
... But Nami wasn't one of the original champions, so it's a reference to something else as well. Possibly simply a reference to pick order's first pick doing dumb things (the songs old enough I don't know what Nami's state was).
In the First Interns at Riot video they showed alpha footage of the game, and it looked pretty drastically different. They showed Singed and Annie (Annie looked more like a teen then), maybe some others.
Point: the answer is whatever the first champion made was, that's the first one locked in, since that champion would be used to test the map/minions/engine.
