Mana regen Questions

Hey Everyone if you could help me out I'm wondering if runes are calculated into a regen items' % increase I can't find a accurate note or anything on this and I want to know if a rune's increase in mana regen is considered "base" regen for something like morello's If anyone can answer this please let me know as fast as possible thank you
