So i have level 5 on my champ ( yasuo ) and i just achieved an S- and an S+ on him. I went to hextech crafting so i could get mastery 6 on yasuo and it says 500 blue essense which i dont have, combine with a champ shard, which i dont have, and it says combine with champion permenant and i have yasuo permanent, if i combine my S- and my S+ with yasuo permanent, will i lose yasuo permenant?

Yes, if you use your Yasuo permanent it is destroyed. It has literally no other use, so just do it. If you need more Blue Essence in the future you can purchase champion shards for 1700 IP apiece.

Just buy two champ shard and convert them 1700 IP each.
