I'd like to think that after destroying all the runes, Ryze becomes a professor to teach young mages about the dangers of runes on runeterra.

If you listen to ryze's quotes he states that after he has gathered all of the world runes he will probably let himself die. He has seen too much and done terrible things, he is tired and once his mission is over he can finally rest.
Ryze is a professor, of Love.
If he destroys all the runes, that not only means he destroys the runes which sustained him, but that also means he would be out of the job as a Professor teaching mages about runes.
