Now before I start I am not in a high rank either mind you, but watching the pros play every year, to reading articles and brushing up, the best advice I can give is pick a role, and pick champions within that role that you enjoy playing and practice, practice PRACTICE. Having an overall grasp of each lane and finding a few champions that work for you within each is not also a bad idea just in case you don't secure that one position you feel you're really good at. At any given time, I always have 2-3 options for each position in mind just in case i don't get who I want. After all in ranked it really is Pick Order > Call order. Hope this helps somewhat!
