Zaun and piltover, though the shadow isles would be nice
Zaun. Revolutionary efforts against the Chem barons, getting to see big ole Urgot as a more established King pin, Viktor's Glorious Evolution advancing forward, potential break down in relations with Piltover that starts a crises for them to boot, which would ofcourse lay the way for that whole Noxian movement on Piltover that they set up earlier, and leave that open door for whenever they go back to advancing Noxus again.
Personally, it would be Bandle City. Would like to know more about the city.
Anything from Piltover/Zaun really, seems kinda lacking in larger ongoing lore.
Ironically Demacia is host to Kayle and Morgana and I'd love to see their stories move forward.
But otherwise, I'd like some follow up on Syndra's situation! She still lacks characterization and an actual goal.
Honestly, any of them. I don't have any strong preference for any faction/region & would prefer to see more of Runeterra progress.
I would also like to see Bandle City get an actual story/plot thread to progress in the 1st place.
Ionia got numerous threads going on due to how large the faction & roster is. Speaking of Ionia, isn't one of the Marvel comic series suppose to center around 
? Hopefully it isn't just a flashback comic.
I would like to see some actual progression on the Freljord front. While the 
comic was nice, it was essentially an origin story that took place in the past.
I'd like Shurima too. In the lore timeline, the last Shurima event to have happened was 5 years ago. Irl, last modern Shurima story was released like....idk 3-4 years ago? Cmon. Not to mention the Universe page gallery is...kinda dissapointing. Unlike other regions, it gives us no clue about Shuriman tools and weaponry or their military (hey it's been 7 years since Bird man and Xerath returned. They had a lot of time to raise armies), nor how some cities look besides the capital and Nashramae (and the capital has 4 images that are almost the same. No depiction showing it how the houses are built from an inside PoV). And on top of that all, unlike every other region, Shurima doesn't even have a dev blog video
I personally would like a 
comic that tells the story of the "Glorious Evolution".
Ionia
While there are a lot of regions that could really use it, Shurima in particular needs its timeline sorted out. Taliyah is depicted in-game as her sixteen-year-old self who's just found out she has to leave Yasuo and go home (I think that's the exact timing?), but she's now canonically 23. Bloodlines seems to have happened well after her in-game appearance, so what else has happened in the past seven years to her and those around her?

all need some kind of lore about their point of existence. Old void was, iirc, just a purple version of our world and something like the upside down from Stranger Things. Kha was the closest to having a story with him and Rengar being rivals, but what happened before that? What's he doing now? Cho has no real place in the lore, and was just the terror of the void, a monster who destroyed and ate everything he encountered, but is he just roaming Icathia? If anything, I'd expect Rek'Sai to be doing that, and she seems to just burrow around and (possibly) creating more mini versions of her, which I guess would help the watchers? Kog'Maw could be a prototype of Vel'Koz, breaking things down with acidic spit and showing the watchers (thought his many eyes) what things are made of, and then Vel'Koz was made to do this in a more scientific manner once the Watchers realized there are smaller particles that make up everything.
The Zaun/Piltover dynamic, I am still a die hard fan of hoping for some Urgot lore and also hoping to see more of Ekko, his interaction with Urgot, Vi, Jinx, Caitlyn.
Also, Bandle City. I REALLY want to know more about them, to see Teemo being implanted fully in canon, etc.
Noxus, I'd like to see Mordekaiser's resurrection, meet the representative of Guile and see the plans of the Black Rose
Shadow Isles. The last update other than the recently released short story was the Yorick update in 2016, and the last new champion in that region was Kalista. Mordekaiser being moved to Noxus is a direction I like, but now there's a notable lack of a major in- game villain at the Isles. There's a ton of evil characters, but there's a lack of a big villain. We've heard about the Ruined King for years and I'd love to finally see him in game. If Mordekaiser returns it could also be through the Mist that allows the other dead to come back. Either way, we need something for the region.
Zaun/Piltover, or shurima.
I'd personally like to see Targon Move on.
They are a large, I mean VERY LARGE center of the lore as is, and more and more of their villains are coming to light, AND they lost their frontline soldier Pantheon, Aspect of War.
Aatrox going on a Siege for Targon's Peak would actually be a nice event, collecting some of Targon's other Enemies (And eventually the other Darkins Rhaast and Varus, with their respective hosts).

Honestly, either Targon so we get to see different interactions of the Aspects&co ( Panth&Soraka, Taric&Panth, Morgana&Soraka, everyone with Aurelion Sol, what both Morgana&Kayle think of AS's imprisonment etc..) , after that? Bilgewater or the Shadow Isle

Ionia, particularly Irelia. Miracle dancer saves her nation, or at least a huge chunk of it? Hurrah, her reward is uncertainty, extremists trying to assassinate her, and a whole mountain of untreated PTSD.
Please tell me that Ionia has small, fuzzy, adorable creatures like kittens or the equivalent. Because I want to see it canonized that Irelia gets a whole bunch of cuddles from them.
