So today I was playing a game of Kassadin and I accidentally killed my Ezreal jg's blue buff and he called me the n word, although with a period between them. I know this seems like a stupid question, but would that period help him avoid the system?
That really depends on how the system was built. You can use something that just looks for the letters next to each other like some companies use, but in that case you end up with situations where you can't type words that are in the game due to chat filters (like Kassadin being blocked because it contains "ass").
Riot isn't normally very keen on showing us how their systems work, if only because they know us too well and don't want us finding every loophole, but the answer to your question is really that it depends.
