Its a legal grey area.
Parody or not, people and companies automatically intellectual property rights over the things that they come up with and market. (in the US at least) If something's found to be too similar or a copy, well then they could be in big trouble. Still so long as the skins are dissimilar enough, riot shouldn't be able to get in trouble. After all Vanilla ice was able to get away with ripping off Queen's under pressure because the beat was i believe 16th of a beat off from the original. Still it could be touch and go for something as distinctive as pikachu.

Pikachu is a Mouse, not a sewer rat. Pikachu is literally based off the animal Pika. Also if Twitch were any pokemon it would be Raticate which is an actual rat.
