Leona and Diana rent lovers, dont add romance to everything. Let it just be arch rivalshipdom (which is better than romance imho). Nasus and Renekton imho have the best relationship. Also Lucian and Thresh, Rengar and Kha,etc.
Since you didn't specify romantic relationship, I say with conviction that Lux and Galio have the best potential relationship. I just want them to be best buds so much.

... Not really, no. There's a lot of relationships in League, most of them far more interesting. The two have never been lovers, so that 'i loved you once now i must slay you' angle that made them interesting previously is gone. Their relationship is now standard 'i know your face and i want to kill you' and we've got dozens of those. Sure, it's pretty obvious that Leona and Diana are gonna end up working together, but they're starting from the position of enemies. Like
