I don't think we can call out a "main" character here. Every champion is an exceptional being and the main character of their own story.

Most influential I would have to call out Swain. Noxus has business basically everywhere and he leads Noxus.

I'd say The Ruined King. He not only effects his region, but also Bilgewater via the Harrowing.

There's no real main characters in League because Riot doesn't want to make players who aren't attached to the champions in question feel left out.

However, in terms of "protagonist" quality, I'd say the following champions have it:

DEMACIA - Garen, Lux, Jarvan IV and Shyvana; maybe Vayne

NOXUS - Katarina, Darius, Riven and Swain

SHURIMA - Taliyah, Sivir, Nasus and Azir

ICATHIA/VOID - Kai'sa, Jax, Kassadin; maybe Malzahar

PILTOVER - Vi, Caitlyn and Ezreal

ZAUN - Janna, Ekko and Jinx; maybe Warwick

IONIA - Shen, Zed, Yasuo; maybe Irelia

FRELJORD - Ashe, Sejuani, Olaf, Ornn; maybe Braum

BILGEWATER - Miss Fortune, Twisted Fate and Graves; maybe Gangplank

BANDLE CITY - Teemo; maybe Tristana

Overall I think Ryze is the most main-est character lore-wise. Simply because A) He has a very interesting, unfinished mission to travel across the entire world to shut down magical threats and hide World Runes and B) Due to his very distant travels, he's come to make a hella lot of relationships with many characters. On his Universe page, many characters like Miss Fortune, Galio and Nasus appear on his 'related' section, and when asked about this (if i recall correctly) a Rioter stated that Ryze meets a lot of different people when he travels to a lot of different places.
