If you are over level 20, runes. Runes runes runes runes runes runes runes. Tier 3 runes will apply to all of your roles and champs and make your lvl 1-3 sooooo much easier, espeically jungle. Right now I suggest flat armor seals, MR/level glyphs, Movement speed quints and penetration marks. Those are the most versatile runes imho as almost any champ can use those stats effectively. Always be sure to try to specialize your rune pages when you can once you develop favorite roles and favorite champs. Good luck!

Create your rune pages, then try to get the champions you want.

Trust me, you want to get runes first. Without runes, you are a SEVERE disadvantage. I used to die a lot after level 20 until I got a full rune page, and then I was able to turn around and hand my enemies a hard smack-down.

Also, champion roster needs to be 14 champions before you can participate in ranked, not including rotation. You also need to be level 30.

I personally would recommend Movement Speed quintessences (good no matter what champion you use) Hybrid Penetration marks (great for hybrid carries and aggressive mages like Ahri) and Scaling Health seals (works great on any team composition.) or possible Scaling Armor (nice overall, but I myself prefer more health)

Scaling AP glyphs, Magic Penetration marks and Spell Vamp quintessences for a more caster-oriented champion.

Attack speed (if your character is more based on auto attacks like Nocturne or Shyvana) or Armor penetration (for caster AD champions like Khazix and Pantheon) along with scaling magic resist glyphs for attack damage champions.

Don't get Flat Runes, by level 5 those are going to be outdone by Scaling Runes. Unless you are a jungler and plan on killing the entire enemy team before the 7-8 minute mark, chances are extremely high your match will go better with scaling. The only exception I find to this is Attack Speed Marks, which are great on champions like Nocturne or Yasuo who greatly benefit from attack speed.
