Mana and health regen runes

Sorry if this is the wrong place to post this question. Let's say I have +6/5sec mana regen runes, does that work at start, level 1? Seems like while my mana is regenerating it only says +3/5s if I'm saying it right. Also, the hextech gunblade healing from damage, does Windspeaker's blessing mastery stack with that bonus? Any help is much appreciated, also, does anyone know when Beta servers open up? Thanks!
